- "Sticky Ends"-restriction enzymes leave behind a specific overhang sequence, giving your DNA fragments directionality.
- When a DNA backbone and DNA insert are digested with the same pair of restriction enzymes (e.g. BamHI and EcoRI) only the cohesive "sticky-ends" can join together.
- This allows full control over the direction of DNA fragment alignment.
- The gaps are then closed by ligation, creating your final DNA fragment.
Double? No Trouble! - How to find the best Double Digest conditions.
Finding the best Double Digest conditions using TWO enzymes IN ONE reaction is no trouble at all. We prepared a chart for you (excerpt is shown below). All you need to do is pick your FastGene® Restriction Enzyme pair and the chart will display the recommended buffers for the reaction.
