Double Digest - Give your DNA directionality.

Double Digest can help you with your cloning experiments. The simultanous DNA cutting with TWO restriction enzymes is frequently used as a powerful cloning tool:

• • • "Sticky Ends"-restriction enzymes leave behind a specific overhang sequence, giving your DNA fragments directionality.
    • When a DNA backbone and DNA insert are digested with the same pair of restriction enzymes (e.g. BamHI and EcoRI) only the cohesive "sticky-ends" can join together.
    • This allows full control over the direction of DNA fragment alignment.
    • The gaps are then closed by ligation, creating your final DNA fragment.
